The research in Nagareddy lab is broadly focused on defining the pathological basis for enhanced atherothrombotic risk to various cardiovascular risk factors such as diabetes, obesity, cigarette smoking, high salt diet, rheumatoid arthritis etc. We specifically focus on identifying the inflammatory cues/ signaling mediators that interact with hematopoietic stem and progenitor cells in the bone marrow that promote systemic inflammation. Our major emphasis is on innate immune cells, particularly the neutrophils that have the ability not only to instigate inflammation but also facilitate the resolution of inflammation. We have identified certain neutrophil-derived alarmin molecules such as S100A8 and S100A9 that activate the NLRP3 inflammasome to initiate inflammation while also retaining the ability to induce anti-inflammatory signaling mechanisms. We use various animal models, human samples, invitro and ex vivo techniques to address our research questions. Our short-term goal is to validate and repurpose certain FDA-approved agents to target chronic inflammation and improve cardiometabolic outcomes. The long-term goal is to harness the wealth of knowledge from our studies and others to develop novel therapeutic strategies to reduce the overall burden of cardiovascular and cardiometabolic diseases. Our research is supported by both federal (NIH) and foundation grants (AHA).
